Hmm hmm hmm.
Well, I'd actually recommend the anime over the manga, first off. Don't get me wrong, I adore the manga, and the anime is pretty much a straight adaption of it. But the thing is, physical comedy is just better when it's actually moving. Same jokes, but funnier when more visceral and with calculated comic timing. Plus, the voice actors are ace all around and that really adds to it.
Both the anime and manga are slow starters, because they have to introduce the supporting cast before they really kick it off and become crazy funny. For the manga, that's about volume 4, and for the anime, that's about fifteen-to-twenty-ish episodes.
So I'd suggest you start with the anime or official manga translations (which are adequate, though if I remember correctly translate the jokes with equivalent substitutes instead of accurate translations,) and as soon as you can get to the stuff by rumbel-subs, who are great because they go above and beyond when it comes to explaining some of the more obscure references (sometimes a bit too beyond, but their hearts are in the right place.) Then when you burn through all of that, switch back to the manga for your Gintama fix. You won't miss anything from going from one to the other, they're literally the same thing except one has movey bits.
